Effects of R 56865 on Na+ Background Current of Ventricular Myocytes



Abstract
Cardiac dysfunction such as myocardial functional failure and ventricular arrhythmia has been largely attributed to intracellular Ca2+ overload. A new cardioprotective drug, R 56865, antagonizes the damaging effects of ischemia-reperfusion injury
including arrhythmia and mechamical deterioration. Although the myocardial protective effects of R 56865 via the inhibition of Na+ influx and consequent prevention of the Ca2+ overload have been reported, the action site of r 56865 is not clearly
invailed. In this study the effects of r 56865 on Na+ background current was investigated in single guinea pig ventricular myocytes. R 56865 decreased this background current effectively at 10E-6M, and its inhibition had dose-dependency. The
blocking
effect of r 96865 on Na| background current may contribute to myocardial protection during ischemia and reperfusion.
